    Abstract:

    T cells are key players in our immune system to fight with cancer cells. However, cancer cells can develop multiple mechanisms to suppress T cell activity to evade immune attack. Signaling suppression and metabolic suppression are the two major parts of immunosuppression, which involves signaling and metabolic checkpoint molecules in T cells. In clinic, blockade antibodies against signaling checkpoints have been successfully applied to treat multiple types of cancer, which highlights the importance of checkpoint biology. In this talk, I will introduce our recent works on the signaling checkpoint PD-1 and the metabolic checkpoint ACAT1. The idea of combination of signaling modulation and metabolic modulation for next-generation cancer immunotherapy will also be discussed.
